Indication

Basal cell carcinoma, locally advanced or metastatic

Dose

Basal cell carcinoma

Oral: 150 mg once daily continuously until disease progression or unacceptable toxicity

Supplied

Available as 150 mg capsules. Capsules should NOT be opened. Store at room temperature in original packaging. Capsules contain lactose monohydrate.

Administration

Take with or without food

Missed Dose

If a dose is missed, patients should be instructed not to take the missed dose but to resume with the next scheduled dose.

Adverse Effects

Common: Diarrhea, Fatigue, Taste disturbances, Weight loss, Muscle spasms, Alopecia.

Less Common: Arthralgia, Stevens-Johnson syndrome, Hepatic injury, Hypersensitivity.

Monitoring Parameters

Baseline before first treatment: CBC & differential, platelets, creatinine. If female of child-bearing potential: pregnancy test (blood) or evidence of hysterectomy.

During treatment: CBC & differential, platelets; if female of childbearing potential: pregnancy test (blood).

As clinically indicated: ALT, bilirubin, sodium, potassium.

Drug Interactions

This drug can impact or be impacted by metabolic enzymes for absorption and excretion. Many drugs can also affect or be affected by these enzymes, so please consult with pharmacy to assess for interactions.
Current drug interaction databases should be consulted for more information.

Clinical Pearls

Avoid in pregnancy and lactation.
Patients should not donate blood while taking vismodegib and for 24 months after the final dose.
Vismodegib is only available through a controlled distribution program called the ERIVEDGE® Pregnancy Prevention Program (EPPP).
